Decontamination



The decontamination operation carried out by embryo transfer (rederivation) is crucial, because it produces animals with a SPF (Specific Pathogen-Free) status from contaminated strains. These decontaminations are carried out using either fresh or frozen embryos.

The TAAM is one of the only laboratories in France to offer this service: its level of expertise has earned it national and international recognition, as a reference training centre in this field.


Decontamination protocol

This procedure involves the transfer of embryos from a contaminated strain into a healthy surrogate mother (SPF). For this, we host 3 to 5 infected males aged over 8 weeks and a dozen female donor embryos between 3 and 5 weeks of age. These females are superovulated by hormonal treatment prior to mating. The next day, the females showing a vaginal plug are isolated.

In parallel, in an isolator (SPF) mature B6CBA F1 females will be mated by vasectomized males. The next day, the females showing a vaginal plug will be isolated and called "pseudopregnant". The oviducts of donor females are collected and embryos are retrieved by injection of medium (M2) into the infundibulum (flushing). The embryos are washed in several successive baths of medium (M2 then M16).

Embryos can be collected and reimplanted from the 2 cell stage to the morula-blastocyst stage. The reimplantation is performed using a binocular microscope and a conventional embryo transfer surgical procedure, into the infundibulum for 2 cell and 4-8 cell stages and into the uterus for the morula-blastocyst stage. Embryo transfer recipients are identified and transferred into an isolator, where they remain until their pups are weaned. At weaning, nursing females are brought to our health control facilty in order to establish a bacteriological, parasitological and virological profile.

After validation of the SPF status by the health service, animals may be, depending on demand, dispatched to animal facilities with SPF status or bred in our own facility
